Firefly Luciferin (Photinus species) D-Luciferin
This is the FREE ACID form and is not soluble in neutral buffers.
Make your buffers alkaline prior to dissolving the d-Luciferin Free Acid, then titrate to your desired pH.
This will facilitate obtaining a precipitate free solution and maximal signal.
Protocol for in vivo use:
make PBS pH=12 and pH = 2
reconstiture 10 mg of coelenterazine in 165 ul of PBS, pH=12 and neutralize it with 165 ul of PBS pH=2.
that would give 30g/L. usually, inject 150 uL of coelenterazine per 25-30 g mouse which gives about 150mg/kg body weight.
